    Polymorphism of fibrinogen Bβ-249C/T gene in smokers with COPD

      目的探讨纤维蛋白原(Fg)Bβ-249C/T基因多态性与慢性阻塞性肺疾病及吸烟的相关性。
      方法慢性阻塞性肺疾病急性加重期住院病人80例作为病例组,同期健康体检者100名作为对照组。采用酶联免疫吸附法检测血浆Fg水平,采用放射免疫法测定IL-8、TNF-α水平;采用聚合酶链反应-限制性片段长度多态性分析技术分析FgBβ-249C/T位点基因多态性,统计基因突变率。
      结果FgBβ-249C/T位点存在基因多态性。2组中吸烟者Fg、IL-8、TNF-α水平和FgBβ-249C/T位点基因型突变比例及CT+TT型Fg水平均明显高于不吸烟者(P < 0.01),且病例组吸烟者Fg、IL-8、TNF-α水平、FgBβ-249C/T位点基因型突变比例及CT+TT型Fg水平均明显高于对照组吸烟者(P < 0.01)。
      结论吸烟与FgBβ-249C/T基因多态性及慢性阻塞性肺疾病存在一定关联性,选择性Fg Bβ-249C/T基因多态性位点的检测有助于临床上对慢性阻塞性肺疾病易患人群的筛查。

       

      Abstract:
      ObjectiveTo investigate the correlation of the polymorphism of fibrinogen(Fg)Bβ-249C/T gene with chronic obstructive pulmonary disease(COPD) and smoking.
      MethodsEighty patients with acute exacerbation chronic obstructive pulmonary disease(AECOPD) and 100 healthy subjects were divided into the case group and control group, respectively.The level of Fg, and levels of IL-8 and TNF-α was detected using the enzyme-linked immunosorbent assay(ELISA) and radioimmunoassay, respectively.The polymorphism of FgBβ-249C/T gene was analyzed using polymerase chain reaction restriction fragment length polymorphism analysis technique, and the mutation rate of gene was calculated.
      ResultsThere was genetic polymorphism in FgBβ-249C/T locus.The levels of Fg, IL-8, TNF-α, gene mutation ratio of FgBβ-249C/T locus and type CT+TT Fg level in smokers were significantly higher than those in nonsmokers in two groups(P < 0.01), and the levels of Fg, IL-8, TNF-α, gene mutation ratio of FgBβ-249C/T locus and type CT+TT Fg level in smokers of case group were significantly higher than those in smokers of control groups(P < 0.01).
      ConclusionsSmoking is associated with the FgBβ-249C/T gene polymorphism and COPD.The selective polymorphism detection of the selective FgBβ-249C/T gene locus may help to screen COPD.
