    Expression and clinical significance of WT1 in patients with different types of myeloproliferative neoplasm

      目的研究不同亚型的骨髓增殖性肿瘤(MPN)病人WT1表达水平差异,总结WT1在MPN中的表达特征,尤其是WT1基因的相对表达量与慢性粒细胞白血病(CML)病情发展的相关性。
      方法运用荧光定量PCR(RQ-PCR)检测56例MPN病人的基因表达类型,以及P210基因、WT1基因的相对表达量,分析WT1在不同基因类型MPN中的表达特点。
      结果CML组WT1阳性率与非CML组比较差异有统计学意义(P < 0.01),CML组的WT1阳性病例中WT1数值与非CML组WT1阳性病例比较差异亦有统计学意义(P < 0.01)。P210+组的WT1阳性率高于非P210+组(P < 0.01),P210+组WT1阳性病例的WT1数值与非P210+组WT1阳性病例比较亦差异有统计学意义(P < 0.01)。WT1和P210基因在CML疾病进程的不同阶段表达差异均有统计学意义(P < 0.01),WT1的数值随着CML进入加速期而明显升高,部分缓解时WT1的数值则明显下降。
      结论MPN病人中WT1的基因表达具有一定的差异性,且其中P210+的CML病人WT1相对表达量可以与P210基因的相对表达量一起作为判断CML进展的指标。

       

      Abstract:
      ObjectiveTo explore the differential expression levels of WT1 in patients with different subtypes of myeloproliferative neoplasm (MPN), and to summarize the expression characteristics of WT1 in MPN, especially the correlation between the relative expression level of the WT1 gene and the progression of chronic myeloid leukemia (CML).
      MethodsFluorescence quantitative PCR (RQ-PCR) was used to detect the gene expression types of 56 MPN patients, as well as the relative expression levels of the P210 gene and WT1 gene.The expression characteristics of WT1 in MPN of different gene types were analyzed.
      ResultsThere was a statistically significant difference in the WT1 positive rate between the CML group and the non-CML group (P < 0.01), and there was also a statistically significant difference in the WT1 value between the CML group and the non-CML group (P < 0.01).The positive rate of WT1 in the P210+ group was higher than that in the non-P210+group (P < 0.01), and the WT1 value of WT1 positive cases in the P210+ group was also significantly different from that in the non-P210+ group (P < 0.01).The expression differences of WT1 and P210 genes at different stages of CML disease progression were statistically significant (P < 0.01).The value of WT1 significantly increased as CML entered the acceleration phase, while the value of WT1 significantly decreased during partial remission.
      ConclusionsWT1 gene expression in MPN has some differences, moreover, the WT1 relative expression level of P210+ CML patients can be used as an index to judge the CML progress together with the relative expression level of P210 gene.
