    Ninjurin2在乳腺癌中的表达及其与预后的关系

    Expression of Ninjurin2 in breast cancer and its relationship with prognosis

      目的探讨神经损伤诱导蛋白2(nerve injury-induced protein 2,Ninjurin2)在乳腺癌中的表达及其与预后的关系。
      方法免疫组织化学染色法检测52例乳腺癌组织及配对癌旁组织中Ninjurin2的表达,分析Ninjurin2表达与乳腺癌临床病理特征及预后的关系。生存分析用Kaplan-Meier法。Western blotting检测乳腺癌细胞系(T47D、SKBR3、HCC1954和MDA-MB231)和正常乳腺上皮细胞系(MCF-10A)中Ninjurin2的表达。用LipofectamineTM 2000试剂盒将Ninjurin2 siRNA序列(沉默组)、阴性对照序列(对照组)分别转入MDA-MB231细胞中,随后用克隆形成实验和CCK8法检测细胞增殖;Transwell实验检测MDA-MB231细胞的侵袭能力;细胞划痕实验检测细胞迁移。
      结果Ninjurin2在乳腺癌组织中高表达率为69.23%(36/52),高于癌旁组织的30.77%(16/52)(P < 0.05)。与MCF-10A细胞比较,T47D、SKBR3、HCC1954和MDA-MB231细胞的Ninjurin2蛋白相对表达量较高(P < 0.05)。Ninjurin2与乳腺癌分期有关(P < 0.05),与年龄、绝经状态、肿瘤大小和淋巴结转移无关(P>0.05)。生存曲线分析结果显示,Ninjurin2高表达病人的无病生存率低于低表达病人(P < 0.05)。与对照组比较,沉默组Ninjurin2蛋白相对表达量、克隆细胞数、细胞培养72 h和96 h时的增殖活力、侵袭细胞数和细胞迁移率均较低(P < 0.05)。
      结论Ninjurin2在乳腺癌细胞及组织中高表达,与乳腺癌临床预后有关。

       

      Abstract:
      ObjectiveTo explore the expression of nerve injury-induced protein 2(Ninjurin2) in breast cancer and its relationship with prognosis.
      MethodsImmunohistochemical staining was used to detect the expression of Ninjurin2 in 52 cases of breast cancer tissues and paired adjacent tissues.The relationship between the expression of Ninjurin2 and the clinicopathological characteristics and prognosis of breast cancer was analyzed.The Kaplan-Meier method was used for survival analysis.The expression of Ninjurin2 in breast cancer cell lines(T47D, SKBR3, HCC1954 and MDA-MB231) and normal breast epithelial cell lines(MCF-10A) was detected by Western blotting.Ninjurin2 siRNA sequence(silencing group) and negative control sequence(control group) were transferred into MDA-MB231 cells with lipofectamineTM 2000 kit, and then cell proliferation was detected by clone formation assay and CCK8 method.The invasive ability of MDA-MB231 cells was detected by Transwell assay, and cell migration was detected by cell scratch test.
      ResultsThe high expression rate of Ninjurin2 in breast cancer tissues was 69.23%(36/52), which was higher than 30.77%(16/52) in adjacent tissues, and the difference was statistically significant(P < 0.05).Compared with MCF-10A cells, the relative expression of Ninjurin2 protein in T47D, SKBR3, HCC1954 and MDA-MB231 cells was higher(P < 0.05).Ninjurin2 was related to breast cancer stage(P < 0.05), but not related to age, menopausal status, tumor size, and lymph node metastasis(P>0.05).The results of survival curve analysis showed that the disease-free survival rate(DFS) of patients with high expression of Ninjurin2 was lower than that of patients with low expression(P < 0.05).Compared with the control group, the relative expression of Ninjurin2 protein, the number of cloned cells, the proliferation activity at 72 h and 96 h of cell culture, the number of invasive cells and cell migration rate in the silence group were lower(P < 0.05).
      ConclusionsNinjurin2 is highly expressed in breast cancer cells and tissues, and is related to the clinical prognosis of breast cancer.
