基于生物信息学数据库分析CDC20基因在膀胱癌中的表达及临床意义

    Expression and clinical significance of CDC20 gene in bladder cancer based on bioinformatics database

      目的探讨膀胱癌中CDC20基因的表达及其临床预后意义。
      方法使用Oncomine数据库和GEPIA数据库分析CDC20基因在膀胱癌中的表达水平。采用Kaplan-Meier Plotter分析CDC20表达水平与膀胱癌病人总体生存时间的关系,采用LinkedOmics数据库分析CDC20基因表达与膀胱癌病人年龄及临床病理特征的相关性,通过STRING数据库绘制CDC20有关蛋白网络图,分析蛋白富集的生理过程。
      结果膀胱癌组织中CDC20基因的表达水平明显高于正常膀胱组织(P < 0.01)。Kaplan-Meier生存分析显示,CDC20高表达的膀胱癌病人生存时间低于CDC20低表达的膀胱癌病人(P < 0.05)。STRING数据库分析显示,与CDC20相关的蛋白有FBXO5、CCNB1、AURKA等,主要富集在细胞周期、卵母细胞减数分裂、孕酮介导的卵母细胞成熟、泛素介导的蛋白水解等生理过程。LinkedOmics数据库分析显示,CDC20的表达与病人年龄、人种均有相关关系(P < 0.05和P < 0.01),与TNM分期及病理分期均无明显相关关系(P>0.05)。
      结论CDC20基因在膀胱癌中高表达,且与膀胱癌病人预后负相关。

       

      Abstract:
      ObjectiveTo investigate the expression of CDC20 gene in bladder cancer and its clinical prognostic significance.
      MethodsThe expression level of CDC20 gene in bladder cancer was analyzed using Oncomine database and GEPIA database.Kaplan-Meier Plotter was used to analyze the relationship between the expression level of CDC20 and overall survival of bladder cancer patients, LinkedOmics database was applied to analyze the correlation between CDC20 gene expression and age, clinicopathological characteristics of bladder cancer patients, and the CDC20-related protein network map was drawn by STRING database to analyze the physiological process of protein enrichment.
      ResultsThe expression level of CDC20 gene in bladder cancer tissue was significantly higher than that in normal bladder tissue(P < 0.01).Kaplan-Meier survival analysis showed that the overall survival of bladder cancer patients with high CDC20 expression was lower than that of bladder cancer patients with low CDC20 expression(P < 0.05).STRING database analysis showed that CDC20-related proteins included FBXO5, CCNB1, AURKA, and so on, which were mainly enriched in physiological processes including cell cycle, oocyte meiosis, progesterone-mediated oocyte maturation, ubiquitin-mediated proteolysis, and so on.LinkedOmics database analysis showed that the expression of CDC20 was correlated with patients' age and race(P < 0.05 and P < 0.01), but was not significantly correlated with TNM stage and pathological stage(P>0.05).
      ConclusionsCDC20 gene is overexpressed in bladder cancer and negatively correlated with the prognosis of bladder cancer patients.
