    Relationship between the expression of occludin in human hepatocellular carcinoma and the genesis and development of hepatocellular carcinoma

    • 摘要: 目的: 探讨紧密连接蛋白occludin表达与人肝细胞癌发生和发展的关系。方法: 运用免疫组化技术检测occludin在人肝细胞癌中的表达,并进行苏木精-伊红(HE)染色观察细胞结构。结果: occludin主要定位于肝细胞膜,且癌旁肝组织的表达阳性率明显高于肝细胞癌(P<0.01).癌旁肝组织occludin表达++~++级强度显著高于肝细胞癌(P<0.005)。occludin在10例分化较好(Ⅰ、Ⅱ级)的肝细胞癌及其癌旁组织中的表达,其阳性率差异无显著性(P>0.05),但33例分化较差(Ⅲ、Ⅳ级)的肝细胞癌中,occludin在癌旁组织中表达的阳性率高于癌组织(P<0.05)。结论: 肝细胞癌与癌旁肝组织相比,occludin表达阳性率及其强度均明显下降,其改变在肝细胞癌的发生发展中可能起重要作用。

       

      Abstract: Objective: To investigate the correlation between expression of occludin in human hepatocellular carcinoma(HCC) and the genesis and development of HCC.Methods: The expression and cellular distribution of occludin were detected by immunohistochemistry.The cellular structure of HCC was observed by hematoxylin-eosin(HE) staining.Results: Immunohistochemistry indicated that occludin was mainly located in the cell junction area.The positive rate of normal liver tissue was higher than that of tumor tissue(P<0.01).The ++-++ grade intensities of non-tumor liver tissue were stronger than those of the tumor tissue(P<0.005).There was no significant difference between the normal tissue and the cancer with better differentiation(Ⅰ,Ⅱ grade) in 10 cases(P>0.05),while the difference was significant between the normal liver tissue and the corresponding HCC tissue with worse differentiation(Ⅲ,Ⅳ grade) in 33 cases(P<0.05).Conclusions: The expression of occludin in hepatocellular carcinoma decreases significantly in both positive rate and intensity compared with that in normal tissue.The decreased expression of occludin may play an important role in the genesis and development of HCC.
