    姜黄素对人胃癌BGC-823细胞体外生长的影响

    Effect of curcumin on the growth of gastric cancer cell BGC-823 ihvitro

    • 摘要: 目的:观察姜黄素(CCM)对人胃癌BGC-823细胞体外生长的影响。方法:将0、30、60、90、120mol/LCCM分别与人胃癌BGC-823细胞共培养24、48h。采用显微镜直接观察BGC-823细胞的形态变化,采用MTT比色法检测BGC-823细胞抑制率,流式细胞术(FCM)检测胃癌细胞周期分布,TUNEL法检测细胞凋亡率。结果:CCM作用后的BGC-823细胞伪足回缩,细胞变小、变圆,瘤巨细胞减少或不见瘤巨细胞。MTT检测显示CCM显著抑制BGC-823细胞的生长,FCM检测提示CCM影响胃癌细胞周期,使其阻滞于G2/M期。TUNEL法表明CCM可以诱导胃癌细胞出现凋亡(p0.05~p0.01)。结论:姜黄素抑制人胃癌BGC-823细胞的增殖,改变其细胞周期分布,诱导胃癌细胞凋亡。

       

      Abstract: To observe the effect of curcumin(CCM)on the growth of gastric cancer cell BGC23. Methods:Humangastric cancer cell BGC23 was cultured with various concentrations of CCM(0,306090 and 120 p,mol/L)for 24 hours and 48hours. The proliferation of BGC23 cell was measured by MTT colorimetric assayand the cell cycle distribution was examined by11owcytometry. Cell apoptosis was determined by TUNEL. Results:The cell's pseudopodia was retracted after CCM treatment the cellsbecame small and round tumor giant cells were reduced or no tumor giant cells. MTT assay showed that CCM was significantly inhibitedthe growth of gastric cancer cells.
