    三氧化二砷对黑素瘤B16细胞黏附和迁移的影响

    The influence of arsenic trioxide on adhesion and migration in melanoma B16 cells

    • 摘要: 目的:了解三氧化二砷(As2O3)对黑素瘤B16细胞迁移能力的影响,评价As2O3抗肿瘤侵袭和转移能力。方法:用Transwell侵袭转移模型评价As2O3抗黑素瘤B16细胞的侵袭和转移作用。MTT法检测B16细胞的黏附能力。流式细胞术检测B16细胞表面黏附分子CD44的表达情况。结果:As2O3可显著抑制B16细胞的迁移和黏附能力(P<0.05~P<0.01),降低细胞表面黏附分子CD44的表达(P<0.01)。结论:As2O3具有抗黑素瘤B16细胞侵袭和转移作用,降低细胞表面黏附分子CD44的表达可能是其机制之一。

       

      Abstract: Objective: To investigate the influence of arsenic trioxide (As2O3) on migration and adhesion capability in melanoma B16 cells,and evaluate the effect of As2O3 on anti-invasion and anti-metastasis ability.Methods: The invasion and migration capacities of melanoma B16 cells were evaluated using Transwell Permeable Supports.The adhesion capacity of melanoma B16 cells was evaluated by MTT assay.The expression of adhesion molecule CD44 was detected by FCM.Results: As2O3 significantly inhibited the adhesion and migration capacities of melanoma B16 cells (P<0.05 to P<0.01) and down-regulated the expression level of CD44 (P<0.01).Conclusions: As2O3 has the effect of antagonizing invasion and metastasis in melanoma B16 cells,the mechanism maybe related to decrease the expression of adhesion molecule CD44.
