    Experimental study of curcumin reversing drug resistance in ovarian cance

    • Objective: To observe the reverse effects of curcumin on Taxol resistance to human ovarian cancer A2780/Taxol cells and analyze the mechanism of the reverse effect of curcumin.Methods: Ovarian cancer cell A2780/Taxol were cultured in vitro,the cells were divided into control group(group A) and experimental groups (different concentrations of curcumin),the cell growth inhibition rate combined with the same concentration of paclitaxel in the two groups were detected by by thiazotyl blue tetrazolium bromide(MTT).The protein expressions of P-glycoprotein(P-gp) and Protein Kinase C-α(PKC-α) in the two groups were detected by Westernblot method.Results: Compared with group A,the cell growth inhibition rates were higher in all experimental groups (P<0.05 to P<0.01).The results of Western blot were shown that the expressions of P-gp and PKC-α in experimental group were significantly lower than those in A group(P<0.01).Conclusions: Curcumin blocked the products of multi-drug resistant protein-1/P-gp and PKC-α in ovarian cancer cells,then increased the toxicity of paclitaxel on the cell and reduced the drug resistance.
