Effect of the neutrophil supernatant on the migration of human umbilical cord mesenchymal stem cells

Abstract
Objective:To investigate the effects of neutrophil supernatant on the migration of human umbilical cord mesenchymal stem cells(UC-MSCs) for providing a theoretical basis in treating the inflammatory disease with MSCs.Methods:The neutrophil cells were isolated form umbilical cord blood,co-cultured with UC-MSCs,and the neutrophil supernatant was extracted.The only UC-MSCs and co-culturing of UC-MSCs with different concentrations of neutrophil supernatant were divided into the control group and experimental group,respectively.The 18h and 40h was set as the culturing time.According to the experimental results of methyl thiazolyl tetrazolium(MTT),the ratios of UC-MSCs to neutrophil supernatant for 1∶0,1∶1 and 1∶50 were investigated.The migration of MSCs at different time-points within 24h were observed using the scratch method.Results:The result of scratch showed that the migration speed of MSCs in 1∶50 cells was significantly quicker than that in 1∶0 and 1∶1 cells.Conclusions:The high concentration of neutrophil supernatant can promote the migration of human umbilical cord MSCs.
