    Effect of IL-10 on the expression of miRNA-125b, and cell proliferation and apoptosis in non-small cell lung cancer

    • Objective:To investigate the interleukin-10(IL-10) on the expression of miRNA-125b,and cell proliferation and apoptosis in non-small cell lung cancer.Methods:The expression level of miRNAs in H460 cells stimulated by IL-10 was detected using real-time fluorescent quantitative PCR.The effects on cell proliferation between IL-10 stimulating and recovery of miRNA-125b expression were compared using CCK8 method.The effects of IL-10 stimulating and recovery of miRNA-125b expression on cell apoptosis were evaluated by flow cytometry.The expressions of miRNA-125b in non-small cell lung cancer and adjacent tissues were verified by real-time fluorescent quantitative PCR.Results:There was different degree expressions of 21 kinds of miRNAs in H460 cells stimulated by IL-10,and the expression decreasing of miRNA-125b was the maximum.After the expression of miRNA-125b recovering,the proliferation and apoptosis of cell stimulated by IL-10 was inhibited and promoted,respectively(P<0.05).The expression of miRNA-125b in non-small cell lung cancer tissue was significantly lower than that in normal tissue(P<0.01).Conclusions:During the cell proliferation induced by IL-10,miRNA-125b can play a role on inhibiting cancer,which is expected to be used as a drug development target in the treatment of non-small cell lung cancer.
