Establishment of osteoarthritis model by medial meniscus partial resection combined with anterior cruciate ligament transecting

    • Objective:To establish the osteoarthritis model by medial meniscus partial resection combined with anterior cruciate ligament transecting, and observe the pathological changes of knee joint in rats. Methods:Twenty female SD rats were implemented with medial meniscus partial resection combined with anterior cruciate ligament transecting in right knee joint, and the left side was set as the control. The whole knee joint was harvested after 1,2,4,6 and 8 weeks of operation, and the knee morphology, joint cartilage structure, synovial ED1-positive cells and X-ray image in knee joints were observed. Results:After 1 week of operation, cartilage cellular swelling, superficial cartilage cells death and decreasing, superficial fibrosis and number of monoclonal antibody ED1 positive cells in synovium increasing were detected. After 4 weeks of operation, the total knee morphological change, bone density under cartilage increasing and synovial hyperfibrosis were observed. Conclusions:The medial meniscus partial resection combined with anterior cruciate ligament transecting can early induce the pathological changes of OA,which is an ideal animal model in osteoarthritis study.
