Effect of long non-coding RNA MALAT1 on the PANC-1 metastasis of pancreatic cancer cell line

Abstract
Objective:To explore the expression of metastasis-associated lung adenocarcinoma transcript 1(MALAT1) long non-coding RNA in pancreatic cancer cells,and its effect on the metastasis of PANC-1.Methods:The expression levels of MALAT1 in 4 pancreatic cancer cell lines were analyzed by real-time PCR.The level of MALAT1 in PANC-1 was down-regulated by small interfering RNA.The abilities of attachment detachment,migration and invasion of PANC-1 were measured using the attachment assay,detachment assay,Transwell migration and invasion assay,respectively.The protein levels of Vimentin,MMP-2 and MMP-9 in PANC-1 were detected by Western blotting.Results:The expression levels of MALAT1 in 4 pancreatic cancer cell lines were different,and which was the highest in PANC-1.After down-regulating the level of MALAT1 in PANC-1,the protein levels of Vimentin,MMP-2 and MMP-9 significantly decreased(P<0.01),and the abilities of attachment detachment,migration and invasion of PANC-1 significantly weakened(P<0.01).Conclusions:Down-regulating the expression of MALAT1 can decrease the levels of Vimentin,MMP-2 and MMP-9,which can attenuate the capacity of PANC-1 metastasis.
