    Study on the miR-655 expression in epithelial ovarian cancer

    • ObjectiveTo investigate the expression level, function and possible molecular mechanism of microRNA (miR)-655 in epithelial ovarian cancer.
      MethodsThe expression levels of miR-655 in 40 cases of epithelial varian cancer tissues and normal tissues were detected using real-time quantitative-PCR, and the relationship between miR-655 expression and clinicopathological features was analyzed.The cell counting Kit-8(CCK-8) and flow cytometry were used to detect the effects of miR-655 on the proliferation and apoptosis of ovarian cancer cells.Western blotting was used to detect the expression of PI3K, AKT and mTOR.
      ResultsThe expression level of miR-655 in epithelial ovarian carcinoma was significantly lower than that in normal tissues(P < 0.01).The differences of the expression levels of miR-655 among patients with different tumor diameter, degree of pathological differentiation and lymph node metastasis were statistically significant(P < 0.05 to P < 0.01), and the differences of the expression levels of miR-655 among patients with different ages and CA-125 were not statistically significant(P>0.05).The transfection of miR-655 mimics could significantly inhibit the proliferation and promote apoptosis of ovarian cancer cells(P < 0.01).The transfection of MiR-655 mimics significantly could significantly inhibit the expression of PI3K, AKT and mTOR in ovarian cancer cells(P < 0.01).
      ConclusionsThe low expression of miR-65 in ovarian cancer tissue is significant, which may be related to the growth and differentiation of ovarian cancer.miR-655 can significantly inhibit the proliferation and induce apoptosis of ovarian cancer cells.miR-655 has a certain value of target therapy in ovarian cancer patients.
