    Expression and clinical significance of miR-432, miR-646 and miR-100 in osteosarcoma cells

    • ObjectiveTo investigate the expression levels and clinical significance of miR-432, miR-646 and miR-100 in osteosarcoma cells.
      MethodsSeventy-two cases of osteosarcoma treated with radical resection and 38 cases of adjacent tissue to osteosarcoma were divided into the osteosarcoma group and control group, respectively.The expression levels of miR-432, miR-646 and miR-100 in two groups were detected using RT-qPCR, and the correlation between the expression levels of miR-432, miR-646 and miR-100, and clinicopathological parameters of patients with osteosarcoma were analyzed.
      ResultsThe expression levels of miR-432, miR-646 and miR-100 in osteosarcoma group were lower than those in osteosarcoma tissues(P < 0.01).The expression levels of miR-432, miR-646, and miR-100 in osteosarcoma tissue cells were not related to the patient's gender, age, tumor diameter, histological type, disease site and TCNR, but the patient's TNM stage was related to the occurrence of lung metastases.The expression levels of miR-432 and miR-100 decreased with the increasing of TNM stage.The expression level of miR-646 in stage Ⅲ was lower than that in stagesⅠand Ⅱ.The expression levels of miR-432, miR-646 and miR-100 in patients with pulmonary metastases were lower than those in patients without pulmonary metastases(P < 0.05 to P < 0.01).The results of Pearson correlation analysis showed that the expression level of miR-432 was positively correlated with the expression levels of miR-646 and miR-100(r=0.76 and 0.79, P < 0.05), while the expression level of miR-646 in tumor patients was positively related to the expression level of miR-100(r=0.81, P < 0.01).
      ConclusionsThe expressions of miR-432, miR-646 and miR-100 are abnormal in osteosarcoma tissue cells, and closely related to the occurrence and development of osteosarcoma, which can provide a basis for the diagnosis and treatment of osteosarcoma in clinic.
