    Methylation and protein expression of RASGRF1 gene in gastric cancer and its clinical significance

    • ObjectiveTo investigate the methylation and protein expression of Ras specific guanylate releasing factor 1(RASGRF1) in gastric cancer and its clinical significance.
      MethodsThe clinical data of 80 patients with gastric cancer were collected.The expression levels of RASGRF1 protein in 80 gastric cancer tissues and corresponding adjacent tissues were detected using immunohistochemistry.The methylation status of RASGRF1 gene was detected by methylation specific PCR.Spearman correlation was used to analyze the correlation of methylation with protein expression levels of RASGRF1 in gastric cancer tissues.The relationship between the methylation status and protein expression levels were observed.
      ResultsThe RASGRF1 protein expression and gene methylation expression were different between gastric cancer tissues and adjacent tissues(P < 0.05).The results of Spearman correlation analysis showed that the RASGRF1 protein expression was negatively correlated with the gene methylation in gastric cancer tissues(P < 0.01).The RASGRF1 protein expression in gastric cancer tissues was not related to the gender, age, tumor diameter, histological differentiation degree and distant metastasis(P>0.05), but was related to tumor stage(P < 0.05).The RASGRF1 gene methylation in gastric cancer tissues was not related to the gender, age, tumor diameter and histological differentiation(P>0.05), but was related to the tumor stage and distant metastasis(P < 0.05 and P < 0.01).
      ConclusionsThere is a negative correlation between RASGRF1 gene methylation and protein expression in gastric cancer tissues.The RASGRF1 gene methylation and protein expression have a certain correlation with tumor stage and distant metastasis.
