Protective effect of rutin on kidney damage induced by high sugar and fat diet in mice

ObjectiveTo study the protective effect of rutin on kidney damage induced by high glucose and fat diet in mice.

MethodsForty male ICR mice were randomly divided into 5 groups, including normal diet group(ND group), high fat diet group(HFD group), rutin protected high-fat diet group(RHFD group), high sugar diet group(HSD group), rutin protected high sugar diet group(RHSD group), 8 rats in each group.After 120 days of continuous feeding, the contents of biochemical indexes such as TC and TG of lipid metabolism were detected.The levels of biochemical indexes such as SOD, GSH-Px and MDA of oxidative stress were measured.The basic morphological structure of kidney tissue was observed by HE staining, and the changes of basement membrane of kidney tissue were observed by PAS staining.

ResultsRutin could significantly reduce the body mass of mice fed with high sugar and fat diet(P < 0.05), increase the activity of antioxidant enzyme GSH-Px in kidney tissue(P < 0.05) and decrease the level of lipid peroxidation product MDA(P < 0.05) to reduce the oxidative stress, which improved the hyperplasia of glomerular epithelial cell and mesangial, the thickening of bowman capsule walland kidney tubular basement membrane, and the vacuolar degeneration of kidney tubular basement membrane.

ConclusionsRutin can effectively improve the kidney damage caused by high sugar and fat diet in mice, which exerts a certain protective effect on the kidney damage caused by high sugar and fat diet in mice.
