ObjectiveTo observe the correlation between the levels of inflammatory factors interleukin (IL) -6, IL-4, IL-8, IL-10, tumor necrosis factor-α (TNF-α) and myeloperoxidase(MPO) in the ulcerative colitis(UC) model rats treated with dandelion polysaccharide.
MethodsThe UC model of rats was established by 2, 4, 6-trinitrobenzenesulfonic acid combined with 50% ethanol solution.The refined dandelion polysaccharide was extracted by water extraction and alcohol precipitation method.Fifty male rats were randomly divided into 5 groups: normal control group, model control group, mesalazine group, dandelion polysaccharide group and mesalazine combined with dandelion polysaccharide treatment group(combined treatment group), with 10 rats in each group.The levels of serum inflammatory factors(IL-6, IL-4, IL-8, IL-10, TNF-α) and the expression of MPO in colon tissue were detected by enzyme-linked immunosorbent assay.
ResultsCompared with the normal control group, the serum levels of IL-6, IL-8 and TNF-α and MPO content in colon tissues were significantly increased, and the levels of IL-4 and IL-10 were decreased in the model control group and dandelion polysaccharide group (P < 0.05), meanwhile, the serum levels of IL-6 and TNF-α were increased and the levels of IL-4 and IL-10 were significantly decreased in the mesalazine group (P < 0.05), and there was no significant difference in the levels of each factor in the combined treatment group (P>0.05).Compared with the model control group, the mesalazine group, dandelion polysaccharide group and combination treatment group had significant reductions in the serum levels of IL-6, IL-8, TNF-α and MPO content in colon tissues, and significant increases in the levels of IL-4 and IL-10 (P < 0.05).Compared with the mesalazine group and the dandelion polysaccharide group, the combined treatment group had significant reductions in the serum levels of IL-6, IL-8, TNF-α and the MPO content in colon tissues, and significant increases in the levels of IL-4 and IL-10 (P < 0.05).The expression levels of IL-6, IL-8 and TNF-α were positively correlated with MPO level, and the levels of IL-4 and IL-10 were negatively correlated with the levels of MPO(P < 0.05).
ConclusionsThe effect of mesalazine combined with dandelion polysaccharide in the treatment of UC is better than that of mesalazine and dandelion polysaccharide alone.The levels of IL-6, IL-8 and TNF-α are positively correlated with the levels of MPO and the progression of inflammation, while the levels of IL-4 and IL-10 are negatively correlated with the levels of MPO and the progression of inflammation.
