    Expression of Ninjurin2 in breast cancer and its relationship with prognosis

    • ObjectiveTo explore the expression of nerve injury-induced protein 2(Ninjurin2) in breast cancer and its relationship with prognosis.
      MethodsImmunohistochemical staining was used to detect the expression of Ninjurin2 in 52 cases of breast cancer tissues and paired adjacent tissues.The relationship between the expression of Ninjurin2 and the clinicopathological characteristics and prognosis of breast cancer was analyzed.The Kaplan-Meier method was used for survival analysis.The expression of Ninjurin2 in breast cancer cell lines(T47D, SKBR3, HCC1954 and MDA-MB231) and normal breast epithelial cell lines(MCF-10A) was detected by Western blotting.Ninjurin2 siRNA sequence(silencing group) and negative control sequence(control group) were transferred into MDA-MB231 cells with lipofectamineTM 2000 kit, and then cell proliferation was detected by clone formation assay and CCK8 method.The invasive ability of MDA-MB231 cells was detected by Transwell assay, and cell migration was detected by cell scratch test.
      ResultsThe high expression rate of Ninjurin2 in breast cancer tissues was 69.23%(36/52), which was higher than 30.77%(16/52) in adjacent tissues, and the difference was statistically significant(P < 0.05).Compared with MCF-10A cells, the relative expression of Ninjurin2 protein in T47D, SKBR3, HCC1954 and MDA-MB231 cells was higher(P < 0.05).Ninjurin2 was related to breast cancer stage(P < 0.05), but not related to age, menopausal status, tumor size, and lymph node metastasis(P>0.05).The results of survival curve analysis showed that the disease-free survival rate(DFS) of patients with high expression of Ninjurin2 was lower than that of patients with low expression(P < 0.05).Compared with the control group, the relative expression of Ninjurin2 protein, the number of cloned cells, the proliferation activity at 72 h and 96 h of cell culture, the number of invasive cells and cell migration rate in the silence group were lower(P < 0.05).
      ConclusionsNinjurin2 is highly expressed in breast cancer cells and tissues, and is related to the clinical prognosis of breast cancer.
