    Study on the relationship between the serum levels of miR-126a and miR-130b and osteoporosis in patients with diabetes mellitus complicated with MS

    • ObjectiveTo investigate the serum levels of micro RNA-126a(miR-126a) and microRNA-130b(miR-130b) in patients with type 2 diabetes mellitus complicated with metabolic syndrome(MS), and their relationships with osteoporosis.
      MethodsA total of 162 patients with type 2 diabetes mellitus were selected.Seventy-two patients with simple diabetes mellitus and 90 patients with diabetes mellitus complicated with MS were divided into the control group and observation group, respectively.The patients with type 2 diabetes mellitus complicated with MS were subdivided into the osteoporosis group(52 cases) and non-osteoporosis group(38 cases).The relative expression levels of serum miR-126a and miR-130b were detected using the real-time quantitative polymerase chain reaction.The laboratory indicators and osteoporosis-related factors levels were detected.The correlation between the expression levels of serum miR-126a and miR-130b and osteoporosis-related factors were analyzed using Pearson method.The multivariate logistic regression analysis was used to analyze the related factors affecting the occurrence of osteoporosis.
      ResultsThe expression levels of serum miR-126a and miR-130b in observation group were significantly higher than those in control group(P < 0.05).The body mass index(BMI), hemoglobin(Hb), osteocalcin, bone mineral density(BMD) and insulin-like growth factor(IGF-1) in osteoporosis group were significantly lower than those in non-osteoporosis group(P < 0.05), and the serum levels of total cholesterol(TC), triglyceride(TG), high density lipoprotein cholesterol(HDL-C), glycosylated hemoglobin(HbA1c), procollagen type Ⅰ amino terminal peptide(P1NP), miR-126a and miR-130b in osteoporosis group significantly increased(P < 0.05).The levels of miR-126a and miR-130b were negatively correlated with BMI, Hb, osteocalcin, BMD and IGF-1(P < 0.01), but positively correlated with TC, TG, HDL-C, HbAlc and PINP(P < 0.01).The results of logistic analysis showed that the BMI, Hb, IGF-1, miR-126a and miR-130b expression levels were the influencing factors of osteoporosis(P < 0.05 to P < 0.01).
      ConclusionsThe expression levels of serum miR-126a and miR-130b increasing in patients with type 2 diabetes mellitus complicated with MS can promote the occurrence of osteoporosis, and whcih may be used as an important auxiliary index to predict the occurrence of osteoporosis.
