    Expression of miR-595 and miR-1246 in serum of ulcerative colitis and its correlation with intestinal flora

    • ObjectiveTo explore the expression of miR-595 and miR-1246 in serum of ulcerative colitis and its correlation with intestinal flora.
      MethodsA total of 160 patients with ulcerative colitis were selected as the observation group.According to the disease activity, the patients were divided into active phase (aUC group) and remission phase (rUC group), with 80 cases in each group.During the same period, 158 healthy people for physical examination were selected as the control group.Real-time fluorescent quantitative PCR (RT-qPCR) was used to detect the expression levels of serum miR-595 and miR-1246.Enzyme-linked immunosorbent assay (ELISA) was used to detect serum tumor necrosis factor-α (TNF-α), interleukin-6 (IL-6), and interleukin-22 (IL-22) levels.Fresh feces of patients and healthy people were collected, and gradient dilution method was used to culture and analyze the flora.
      ResultsThe expression levels of miR-595, miR-1246, TNF-α, IL-6 and IL-22 in the aUC and rUC group were significantly higher than those in the control group (P < 0.05), and which in the aUC group were higher than those in the rUC group (P < 0.05).The number of lactobacillus and bifidobacteria in the aUC group and rUC group were lower than those in the control group (P < 0.05), and which in the aUC group were lower than those in the rUC group (P < 0.05).The number of enterococcus and Escherichia coli in aUC group was significantly higher than that in control group and rUC group (P < 0.05).Pearson correlation analysis showed that the expression levels of miR-595, miR-1246, TNF-α, IL-6 and IL-22 were negatively correlated with the number of lactobacilli and bifidobacteria (P < 0.05), and positively correlated with the number of enterococci and Escherichia coli (P < 0.05).
      ConclusionsThe expression of miR-595 and miR-1246 in the serum of ulcerative colitis is significantly up-regulated, and closely related to the number intestinal flora.
