Abstract
Objective: To observe the expression of survivin and caspase-3 in brain glioma and effect of survivin antisense oligodexynucleotides(ASODN) on expression of survivin and caspase-3 in U251 cells,and explore the relation of the expression of survivin and caspase-3 to the formation and clinical prognosis of brain glioma.Methods: Survivin and caspase-3 protein were detected by immounohistochemistry of glioma.The glioma cells,U251 were transferred with survivin ASODN(100 nmol/L,300 nmol/L,500 nmol/L),expression of surviving and caspase-3 of tumor cells were measured by RT-PCR and immounohistochemistry.Results: The positive rates of survivn and caspase-3 in glioma were 69.6% and 50.7%,respectively,compared with health control,the differences were significant(P<0.05-P<0.01).Survivin ASODN induced downregulation of the survivin mRNA in a time and dose-dependent manner,survivin ASODN also induced downregulation of the survivin protein and upregulation of the caspase-3 protein.Conclusions: Survivn upregulated and caspase-3 downregulated in glioma.Survivin ASODN can downregulate the expression of survivin mRNA and protein and upregulate the expression of caspase-3 protein.
