Arsenic trioxide induced apoptosis in human bladder cancer BIU-87 cell line and its influence on cell cycle

Abstract
Objective: To observe the effects of arsenic trioxide (As2O3) on the cell growth,cell cycle and apoptosis of human bladder cancer BIU-87 cells line.Methods: CellTiter 96 Aqueous One was used to determine the cell proliferation.Morphological changes of BIU-87 cells were observed by fluorescence staining of Hoechst 33258.Cell cycle and apoptosis were analyzed by flow cytometry (FCM).Results: There was obviously a concentration-dependent relationship between As2O3 and the inhibition of BIU-87 cell proliferation(P<0.01).IC 50 was 28.92 μmol/L.Morphological observation by fluorescence staining indicated that As2O3 induced apoptosis of BIU-87 cells.FCM demonstrated that the number of cells in G0/G1 phase was decreased,while that in S phase were increased after treated with As2O3,and apoptosis rate was 47.37%.Conclusions: The results suggest that As2O3 can inhibit human bladder cancer cells BIU-87 growth by means of cell proliferation inhibition,S phase arrest and induction of apoptosis.
