Expression and significance of toll-like receptor 4 and tumor necrosis factory in rat colon tissue with ulcerative colitis

Abstract
Objective:To explore the expression of toll-like receptor 4(TLR4) and tumor necrosis facor-(TNF-) and analyse their relationships in rat colon tissue with ulcerative colitis(UC),and investigate the significance of TLR4 signaling pathway in the pathogenesis of UC.Methods:Eighty SD rats were randomly divided into control group(20 rats) and model group(60 rats) which were made model using trinitrobenzene sulfonic acid and ethanol.The morphology and histological changes in the model group were observed.The expression of TLR4 and TNF- in colonic mucosa in two groups were measured using immunohistochemistry.Results:The successful rate of making model was 97%.The expression rates of TLR4 and TNF- protein in UC colon epithelium cells and inflammatory cells were 84.5% and 89.7% respectively,which were higher than that of the control group of 30.0% and 20.0%(P0.01).The expression of TLR4 and TNF- in colonic mucosa in the model group were significantly enhanced with histological grade increasing and its were a positive relationship(P0.01).Conclusions:The expressions of TLR4 and TNF- have some relation to the occurrence and development of UC.
