Impact of estrogen on the expressions of cyclooxygenase and proliferating cell nuclear antigen in cervical cancer Hela cells

Objective:To investigate the regulation of 17 estradiol(E=)on the expressions of cyclooxygenase(COX 2)as well asproliferating cell nuclear antigen(PCNA)in cervical cancer Hela cells. Methods:Hela cells were treated with 0. 1% alcohol(vehicle)or E= or E=+etoricoxib for 24 hours followed by the examination of COX or PCNA expressions using immunocytochemistry Westernblot and reverse transcriptase-polymerise chain reaction(RTCR)assay. Results:The level of COX 2 and PCNA protein as well asmRNA in cells of E= group was significantly increased compared with control. The level of PCNA in cells of E=+etoricoxib group wasnotably inhibited relative to E=,but still higher than control. Conclusions:E= can induce COX expression in Hela cells therebyupregulating PCNA level
