    Comparison of Mycobacterium tuberculosis heat-resistant antigens and phosphoantigens on activation and proliferation of human peripheral T cells

    • Objective:To compare the characteristics of Mycobacterium tuberculosis heat-resistant antigens(Mtb-HAg) and a kind of phosphoantigens,and HDMAPP on activation and proliferation of human peripheral blood T cells.Methods:The peripheral blood mononuclear cells(PBMC)(1106/ml) from healthy adults were cultured with Mtb-HAg(5 g/ml) of HDMAPP(210-9mol/ml),and IL-2(50 u/ml)added to maintain cell proliferation.The group in which only IL-2 added was as control.After cultured for 10 days,T cells were labelled with anti-human-CD3-FITC and anti-human-TCR-PE,and detected by flow cytometry.Results:After amplification,the proportion of T cells in Mtb-HAg activated group was significantly lower than that in HDMAPP group(P0.05).The proportion of T cells in these two groups were significantly higher than that in IL-2 control group(P0.01).Each individual had a linear correlation with the two kinds of antigen stimulated amplification of T cells(R2= 0.855 1,P0.01).Conclusions:Both Mtb-HAg and HDMAPP can preferentially activate human peripheral T cells,and HDMAPP has a stronger activity.
