Effect of tea polyphenols on glyphosate induced sertoli cell injury in mouse

Abstract
Objective:To investigate the effect of tea polyphenols(TP)on glyphosate induced mouse sertoli cell(SC)injury.Methods:The mouse sertoli cell was cultured and identified by oil red O.The cells were divided randomly into control group,glyphosate model group(the SC was treated with glyphosate at 90 g/ml)and TP pretreatment group(the SC was pretreated with 40,80,160 g/ml TP for 12 hours,then treated with glyphosate for 24 hours respectively).The cell viability was measured by metylthiazdyltetrazolium method,lactate dehydrogenase(LDH)activity was determined by colorimetric method,and the cell apoptosis was observed by TUNEL method.Results:In contrast to control group,the cell survival rate was decreased,LDH activity and cell apoptosis ratio were increased in glyphosate model group(P0.01).In TP pretreatment at 40 g/ml and 80g/ml groups,the cell survival rate was significantly improved,LDH release and cell apoptosis ratio were decreased(P0.01).In TP pretreatment at 160 g/ml group,there were no significant changes than that in glyphosate model group(P0.05).Conclusions:The appropriate concentrations of TP at 40 g/ml and 80 g/ml can protect sertoli cell against glyphosate induced injury.
