Expression of high risk type human papillomavirus in the tissues and peripheral blood of cervical carcinomas

    • Objective: To detect expression of human papillomavirus(HPV) type 16 and 18 in carcinoma tissues and peripheral blood of patients with cervical carcinomas, and to explore the relationship between HPV infection and cervical carcinomas,as well as its expression in tumour micrometastase. Methods: Real-time fluorescence quantitative PCR(FQ-PCR) and reverse transcription polymerase chain reaction(RT-PCR) were used to detect HPV in 24 cases of tissues and 48 samples of peripheral blood of cervical carcinomas, and 20 cases of tissues and 20 smples of peripheral blood of chronic cervicitis. Results: In the 24 tissues of cervical carcinomas(Ⅰ-Ⅱb),the positive rate of HPV DNA type 16 was 66.7%;the positive rate of HPV DNA type 18 was 29.2%;the positive rate of both positive HPV DNA 16 and HPV DNA 18 was 8.0%.In the 48 samples of peripheral blood cervical carcinomas, the positive rate of HPV mRNA type 16 was 2.1%.In the 20 chronic cervicitis tissues, the positive rate of HPV DNA type 16 was 15.0% and HPV DNA type 18 was negative. Both HPV type 16 and 18 of peripheral blood were negative. The difference was markedly significant between cervical carcinomas and chronic cervicitis(P<0.005-P<0.05).Conclusions: The quantitation of HPV is highly associated with cervical carcinomas.Detection of HPV mRNA in the peripheral blood of cervical carcinomas is low,and its clinical significance needs further evaluation.
