Abstract
Objective:To investigate the effect of angiotensin Ⅱ(AngⅡ) on fibronectin(FN) biosynthesis on human renal tubular epithelial cells.Methods:Cultured human renal tubular epithelial cells were treated with AngⅡ at the concentrations of 10-6 mol/L for 6,12,24,48,72 and 96 hours;and the cells were treated with AngⅡ at the concentrations of 10-8,10-7 and 10-6 mol/L for 96 hours.The FN expression in renal tubular cells was determined by both Western blot and immunofluorescence.Results:Fibronectin synthesis was increased by AngⅡ at a concentration of 10-6 mol/L in the renal tubular epithelial cells.The expression of FN started to increase after 12 hours,and reached the peak after 72 hours.The synthesis of FN was the most obvious at a concentration of 10-6 mol/L,and the following was at 10-7 and 10-8 mol/L.Conclusions:It suggests that AngⅡ stimulates FN secretion of human renal tubular epithelial cells in a dose and time-dependent manner.
