Abstract
Objective: To investigate the effect of etoposide(VP-16),in combination with oligodeoxynucleotides on lung adenocarcinoma cells.Methods: Etoposide in combination with telomerase 11-base oligodeoxynucleotides werer transfected into SPC-A1 cell line by lipofection.The multiparameters such as cell growth inhibition rate,cell cycle and apoptasis rate,and telomerase activity were detected by MTT,FCM,and PCR-ELISA,respectively.Results: Etoposide inhibited cell growth(with an inhibition rate of 18%),decreased telomerase activity and induced cell apoptosis.ASODN+VP-16 and SODN+VP-16 inhibit the growth of SPC-A1 cell line and rate are 43.5% and 41.0%,repectively(P>0.05).The difference was significant when compared with VP-16(P<0.01).Conclusions: Chemotherapeutant in combination with short chain oligodeoxynucleotides is more effective than used alone in inhibiting lung cancer and the side effect can be greatly reduced.
